Structured lattice light-sheet microscopy for high-temporal-resolution imaging of axonal action potential propagation
The work will include the development of the optical illumination and detection architecture, the integration of metasurface components for the generation and control of the structured illumination pattern, and the design of the system to ensure compatibility and integration with micro-nano-fluidic devices for sample confinement and controlled positioning. Particular attention will be devoted to the development of strategies for the controlled translation and modulation of the illumination pattern and to the synchronization of different illumination configurations with high-speed image acquisition. In parallel, computational image reconstruction procedures will be developed and optimized using acquisitions obtained with different positions or configurations of the illumination pattern, with the aim of increasing spatial resolution while preserving high temporal resolution
