Patient-derived secretome mapping to track tumor microenvironment reprogramming by targeting the nuclear lamina in Glioblastoma
The appointee will support secretome collection from glioblastoma tissue obtained through surgical resection and contribute to isolating and culturing patient-derived GSC. Thanks to experience in gene transduction protocols and clonal selection, they will also help generate genetically modified GSC models for the project's pharmacological approach. They will take part in multiplex secretome profiling and its validation, as well as in drug treatments aimed at inducing prelamin A accumulation and oxidative stress; expertise in nuclear phosphoinositides will help explore this signaling as a mechanism complementary to lamina alterations. They will be responsible for statistical processing of secretome datasets, supporting a translational response model. Donor tissues from the Anatomy Center will serve as a control brain reference. Finally, they must document the work, follow good laboratory practice, and collaborate with the involved groups.
